Travel Interstate 40 East from Knoxville until Exit 421 on the left for I-81 North, toward Bristol. Follow I-81 North to Hwy 26E South, toward Johnson City. From 26E South, take Exit 31 for Hwy 321 South, toward Elizabethton. Stay on 321 around Watauga Lake (this road is curvy -- heed the warning signs for curves). Follow Hwy 321 North into Boone, NC. Note that Hwy 321 North will merge with Hwy 421 South just outside Boone and requires a right hand turn.

Turn right at the first stoplight in Boone, marking the Hwy 105 Bypass, and follow until it ends at a stoplight at Hwy 105. Turn left in the right hand left turning lane onto Hwy 105 North, then turn right onto Poplar Grove Road just past the Tomato Shack on the right. Follow Poplar Grove Road for 3.8 miles until it ends at Shulls Mill Road. Turn left onto Shulls Mill Road. Entrance to the Inn at Yonahlossee is 300 yards ahead on the right.
